CoNi alloy nanoparticles for cancer theranostics: synthesis, physical characterization, in vitro and in vivo studies

Abstract Nanomaterials are attracting increasing interest in many biomedical fields, including the fight against cancer. In this context, we successfully synthesized CoNi alloy nanoparticles (NPs) by a simple polyol process. The magnetic characteristics of products were measured vibration sample magnometry, which revealed that samples have soft ferromagnetic behavior. microstructure and morphology inspected X-ray diffraction scanning electron microscopy, respectively. Human cancer cells derived from breast (MCF7) oral cavity (C152) normal human umbilical vein endothelial (HUVECs) treated with concentrations NPs, their cytotoxic effect was via MTT lactate dehydrogenase (LDH) leakage assays. We found treatments using 12.5 to 400 µg/mL Co0.5Ni0.5, Co0.6Ni0.4, Co0.4Ni0.6 NPs associated significant concentration-dependent toxicity toward such cell lines profoundly enhanced LDH following 48 h exposure ( P < 0.05 compared untreated cells). Besides, NP dose 6.25 did not affect survival HUVECs while leading marked death MCF7 C152 cells. vivo experiments rats done investigate biochemical histopathological changes over three weeks, intraperitoneal administration (100 mg/kg). As controls, caused elevations aspartate aminotransferase, alanine blood urea nitrogen, serum creatinine, catalase activity, superoxide dismutase, liver malondialdehyde levels. Also, Co0.6Ni0.4 showed more severe kidney. Our findings represent an essential step developing theranostic nanoplatforms for selective treatment.
